Inhibitory effect of Diospyroskaki, Chenogdo flat persimmon on cell differentiation and lipid metabolism-related protein in 3T3-L1 preadipocyte (pilot study)

Abstract
Objective : In this study, we investigated the preliminary anti-obesity capacity of water extract from Bansi (Diospyros kaki. CBS), seedless persimmon produced in Cheongdoin using 3T3-L1 preadipocyte.

Methods : The cellular toxicity of CBS extract (CBSE) was measured by MTT assay. And antiobesity-related activities were evaluated via Oil-Red-O staining for monitoring inhibitions on adipogenesis and lipid accumulations, and western blot for identifying expression of differentiation factor (AMPK, ACC, PPAR¥ã, C/EBP¥á) in 3T3-L1 cells.

Results : Lipid accumulation of CBSE-treated group significantly decreased in a dose dependent manner compare to that of vehicle-treated control group, which is 83.7% and 72.7% at 1§·/§¢and 2§·/§¢ CBSE concentration, respectively. And pAMPK and pACC protein expression of 2mg/ml CBSE-treated groups were higher than vehicle-treated control group. The expression levels of PPAR¥ã and C/EBP¥á in 2§·/§¢ CBSE-treated groups were lower than control group.

Conclusion : Therefore, these results suggest that the CBSE could be used as a new material for functional food and drink to improve overweight or obesity.
